The training session was led by Profe Ortega and focussed on fitness work.
After Atleti's pre-season trip to Los Angeles de San Rafael was ended earlier than planned because of a case in the camp, Simeone and everyone at the club had a PCR test on Friday morning.
The results came back on Saturday morning and weren't conclusive, so the 50-year-old had another test and this one has returned a positive result.
He is asymptomatic, as announced by the club on Saturday night.
Now, the coach must self-isolate.
Atletico Madrid don't have a competitive fixture for two more weeks, since they won't start their 2020/21 LaLiga Santander campaign until the third matchday.
“The first team and the staff were subjected to tests on Friday, as soon as they returned from the training camp in Los Angeles de San Rafael after having revealed a positive member of the group in the previous tests on Thursday,” a statement by the club wrote.
“The analysis on the new tests has determined that our coach, Diego Pablo Simeone, gave a positive result for COVID-19. He has no symptoms and is in home isolation."
