Zinedine Zidane had rested Karim Benzema while likes of Cristano Ronaldo, Casemiro, Toni Kroos were given breaks ahead of second ChampionsLeague clash against Paris Saint-Germain.
Gerrard Moreno scored the only goal for Espanyol in the 90th minutes in a game that saw Gareth Bale lead the attack.
The former Spurs player was unable to convert his chances in the game.
